Sanger Sequencing is the gold-standard DNA sequencing technique to determine the precise order of nucleotides base (A-adenine, G-guanine, C-cytosine, T-thymine) in the DNA. This is based on selective incorporation of the chain-terminating dideoxynucleotides by DNA polymerase during in vitro DNA replication. As the result, variable length DNA fragments are synthesized and separated by electrophoresis to reveal the sequence of the DNA. This technology can be used to detect variants (SNPs), mutations and methylation events etc.
